General description
NovaTaq Hot Start DNA Polymerase is a chemically modified Taq DNA Polymerase, which is inactive at room temperature. This polymerase is a derived recombinant form of Thermus aquaticus DNA polymerase. The enzyme exhibits a 5′→3′ DNA polymerase activity and lacks 3′→5′ exonuclease activity. The enzyme provides improved specificity when compared to standard Taq polymerase and can eliminate the presence of nonspecific amplification such as primer-dimers and mis-primed products. The enzyme must be activated by heat treatment (7–10 min at 95°C), after which thermal cycling can proceed. NovaTaq Hot Start DNA Polymerase generates PCR products with 3′-dA overhangs, suitable for cloning with Novagen® Perfectly Blunt®, Acceptor, and LIC Vector Kits. Unit Definition
One unit is defined as the amount of enzyme that will catalyze the incorporation of 10 nmol of dNTP into acid-insoluble form in 30 min at 72°C, in a reaction containing 25 mM TAPS (tris-[hydroxymethyl]-methyl-amino-propane-sulfonic acid, sodium salt), pH 9.3 at 25°C, 50 mM KCl, 2 mM MgCl₂, 1 mM 2-mercaptoethanol, 0.2 mM dATP, dGTP, and dTTP, 0.1 µM [A-³²P]dCTP, and 12.5 µg activated salmon sperm DNA in a volume of 50 µl.
